Zusammenfassung
In der vorliegenden Arbeit wird über die zeitlichen Relationen von Virus-DNS-Synthese einerseits und Riesenzellbildung, Proteinsynthese sowie virusbedingter Hemmung der Zell-RNS-Synthese andererseits berichtet. Die Ergebnisse wurden autoradiographisch durch Einbau von Tritium-markiertem Thymidin, Uridin und Aminosäuren in Kombination mit zeitlich abgestuften Gaben von Actinomycin C, Actidion oder pFPA gewonnen.
Bei den Untersuchungen hat sich herausgestellt, daß eine synchrone Transcription innerhalb der ersten 2 Stunden p.i. und eine synchrone Translation 2 bis 3 Stunden p.i. als Voraussetzung für alle 3 Prozesse anzusehen ist. Virus-DNS-Synthese und Riesenzellbildung werden gleichmäßig erst bei hohen Konzentrationen von pFPA gehemmt, während die Neubildung infektiöser Viria schon bei viel geringeren Konzentrationen von pFPA gehemmt wird. Gleichzeitig mit der Riesenzellbildung beginnt auch die Volumenzunahme der Kerne. Die Riesenzellbildung geht nicht mit ausgeprägten Störungen der Proteinsynthese einher. Vielmehr ist die Riesenzellbildung offenbar eng mit der Frühproteinphase der Virus-synthese verknüpft und nicht mit der Entstehung kompletter, infektiöser Teilchen. Die Ergebnisse wurden im Vergleich zur Riesenzellbildung bei Myxoviren diskutiert.
Summary
The present work concerns the time relationships of viral DNA synthesis on one hand, and of giant cell formation, protein synthesis and virus induced inhibition of cell RNA synthesis on the other hand. The results were obtained with autoradiographic techniques after incorporation of3H labeled thymidine, uridine or amino acids combined with timely restricted inhibitory action of actinomycin C, actidione or pFPA.
The results of these experiments have shown that all three processes were best seen as resulting from a synchronous transcription within the first two hours after infection and a synchronous translation 2–3 hours after infection. The synthesis of viral DNA as well as the formation of giant cells are identically inhibited by high concentrations of pFPA only, whereas the formation of new virions is inhibited at much lower concentrations of pFPA. The volume increase of the nucleus is simultaneous to the giant cell formation. The latter is not linked with remarkable alterations of protein synthesis. It is more openly linked to the synthesis of early proteins than with the production of new virions. The results are discussed in relation with giant cell formation induced by myxoviruses.
