Zusammenfassung
Bei der Vermehrung von Rubellavirus in BHK21-Zellen wird3H-Uridin in Partikel eingebaut, die sich bei der Sedimentation im Saccharosegradienten und bei der Dichtebestimmung im CsCl-Gradienten wie hämagglutinierende Partikel verhalten. Hitzedenaturierung des markierten Materials setzt RNase-empfindliche Radioaktivität frei. Nach Behandlung des Materials mit Tween 80 und Diäthyläther lassen sich Hämagglutinin und3H-markiertes, nukleinsäurehaltiges Material durch Dichtegradientenzentrifugation trennen. Die Befunde stützen die Auffassung, daß das Rubellavirus ein RNS-Virus ist.
Summary
During the propagation of rubella virus in BHK-cells3H-uridine is incorporated in particles which can not be distinguished from hemagglutinating particles by sedimentation in sucrose or by density gradient centrifugation in CsCl. Heat denaturation of the labelled product is followed by a release of radioactivity which is sensitive to RNase. After treatment of this material with Tween 80 and diethyl ether the hemagglutinin can be separated from a3H-labelled, nucleic acid containing material by density gradient centrifugation. These findings support the conception that rubella virus is a RNA-virus.
