Summary
Prostaglandins are hormone-like substances, which are synthetized in mammals from C20-fatty acids with 3–5 double bonds. Recently it was shown that these substances inhibit the activation of lipolysis in fatty tissue. We have investigated the effect of 0.1μg/ml PGE1 (prostaglandin E1) on the carbohydrate and fat metabolism of isolated epididymal tissue in rats, utilizing glucose-containing buffers with and without addition of 500μU/ml insulin, 0.1μg/ml adrenaline and 25μg/ml human growth hormone. At the above mentioned concentration, PGE1 influenced the basal lipolysis (release of glycerol) to a greater extent than insulin. — The growth hormone- and adrenaline-induced activation of glycerol liberation from fatty acids was decreased by PGE1. The basal oxidation of 1-14C-glucose to14CO2 was stimulated by about 70% through the action of PGE1. This stimulation amounted to nearly one-tenth of the effect of 500μU/ml of insulin. The influence of PGE1 on glucose oxidation could be significantly increased by the addition of adrenaline and growth hormone. Glucose oxidation stimulated by PGE1 amounted to one-third of the stimulation produced by insulin, and corresponded to the effect, of 0.1μg/ml of adrenaline and 25μg/ml of growth hormone. PGE1 only intensified the increase in glucose uptake caused by adrenaline. The effect of growth hormone was not influenced. Addition of PGE1 to the incubation media raised by almost 80% the incorporation of 1-14C-glucose into the lipids of the epididymal fatty tissue; this corresponded to approximately 10% of the effect of insulin, 40% of the effect of adrenaline and 80% that of growth hormone. The effect of the last two hormones was slightly stimulated by the simultaneous addition of PGE1 to the incubation media. These experiments indicate that PGE1 influences both glucose and lipid metabolism in epididymal fatty tissue. It is proposed that the stimulating effect of PGE1 on glucose utilization is a consequence of regulatory changes in cellular metabolism and is closely related to antilipolytic activity.

Les prostaglandines sont des substances dont les actions sont semblables à celles des hormones etqui sont synthétisées dans l'organisme des mammifères à partir d'acides gras à 20 atomes de carbone avec 3–5 doubles liaisons. On a décrit récemment que ces substances inhibent l'activation de la lipolyse au niveau du tissu adipeux. — Nous avons étudié l'action de la prostaglandine E1 (PGE1), 0.1μg/ml, sur le métabolisme des lipides et des hydrates de carbone dans le tissu adipeux épididymaire du rat. Les tissus ont été incubés dans une solution tampon contenant du glucose, avec ou sans insuline, 500μU/ml, adrénaline, 0.1,⧎/ml, ou hormone somatotrope, 25,μg/ml. Nous avons observé que la lipolyse basale est affectée davantage par la PGE1 que par l'insuline, aux concentrations indiquées, et que la PGE1 diminue fortement l'activation de la libération de glycérol induite par la présence de somatotropine ou d'adrénaline. En présence de PGE1 l'oxydation basale de glucose-1-C14 est augmentée de 70%, ce qui représente un effet de dix fois inférieur à celui de l'insuline. L'action de la PGE1 sur l'oxydation du glucose est augmentée en présence d'adrénaline ou d'hormone somatotrope. La PGE1 augmente également l'utilisation du glucose bien que son effet ne représente environ qu'un tiers de celui de l'insuline; cet effet est approximativement équivalent de celui résultant de la présence de 0.1μg/ml d'adrénaline ou de 25 mg/ml d'hormone somatotrope. L'augmentation de l'utilisation du glucose résultant de la présence de l'adrénaline est renforcée par la PGE1; l'action de l'hormone somatotrope n'est pas influencée par sa présence. La PGE1 augmente de 80% l'incorporation de glucose-1-C14 dans les lipides épididymaires du tissu adipeux, un effet qui, de nouveau, représente environ un dixième de celui de l'insuline et environ 40 à 80% de l'action de l'adrénaline ou de l'hormone somatotrope. L'action de ces deux hormones est augmentée par l'addition simultanée de la PGE1 au milieu d'incubation. —Ilsemblerait donc que la PGE1 n'influence pas seulement lemétabolisme du glucose mais également celui des lipides du tissu adipeux épididymaire de rat. Il est probable que la stimulation du métabolisme du glucose par la PGE1 est le résultat des réajustements du métabolisme cellulaire par suite de son action antilipolytique.
Zusammenfassung
Prostaglandine sind hormonartige Wirkstoffe, die im Säugetierorganismus aus C20-Fettsäuren mit 3–5 Doppelbindungen synthetisiert werden. Vor kurzem wurde nachgewiesen, daß diese Verbindungen die Freisetzung von unveresterten Fettsäuren und Glycerin aus dem Fettgewebe hemmen. — In eigenen Versuchen wurde die Wirkung von 0.1 ⧎/ml Prostaglandin E1 (PGE1) auf den Kohlenhydrat- und Fettstoffweehsel des isolierten epididymalen Fettgewebes der Ratte in glucosehaltiger Pufferlösung mit und ohne Zusatz von 500μE/ml Insulin, 0.1μ/ml Adrenalin und 25⧎g/ml menschlichem Wachstumshormon geprüft. — In den angegebenen Konzentrationen ist der Einfluß von PGE1 auf die basale Lipolyse (Glycerinfreisetzung) etwas größer als die Insulinwirkung. Die durch Wachstumshormon und Adrenalin induzierte Aktivierung der Glycerinfreisetzung aus dem Fettgewebe wird durch PGE1 stark herabgesetzt. — Die basale Oxydation von 1-14C-Glucose zu14CO2 wird durch PGE1 um annähernd 70% gesteigert. Im Vergleich zur Wirkung von 500μE/ml Insulin ist der PGE1-Effekt um fast eine Zehnerpotenz geringer. Der Einfluß von PGE1 auf die Glucoseoxydation läßt sich durch Zusatz von Adrenalin und Wachstumshormon signifikant erhöhen. — Die Glucoseaufnahme erfährt durch PGE1 eine Steigerung, die annähernd 1/3 der Insulin Wirkung ausmacht und die in etwa der Wirkung von 0.1 ⧎/ml Adrenalin und 25 ⧎/ml Wachstumshormon entspricht. PGE1 verstärkt lediglich die durch Adrenalin verursachte Zunahme der Glucoseaufnahme. Der Effekt des Wachstumshormons wird nicht beeinflußt. — Durch Zusatz von PGE1 zum Inkubationsmedium wird der Einbau von 1-14-Glucose in die Lipide des epididymalen Rattenfettgewebes um fast 80% verstärkt. Das entspricht etwa 10% der Insulinwirkung und 40% bzw. 80% der Wirkung von Adrenalin bzw. Wachstumshormon. Die Effekte der beiden letztgenannten Hormone werden durch gleichzeitige Zugabe von PGE1 zur Inkubationsflüssigkeit gering gesteigert.— Aus den Untersuchungen geht hervor, daß PGE1 sowohl in den Glucose- als auch in den Fettstoffwechsel des epididymalen Rattsnfettgewebes eingreift. Es wird vermutet, daß der stimulierende Effekt von PGE1 auf die Glucoseutilisation Folge einer regulatorischen Umstellung im Zellstoffwechsel ist und mit seiner antilipolytischen Wirkung zusammenhängt.
