Microdetermination ofα-amino-acids by direct titration with copper(II) sulphate solution

Summary

A simple, rapid, accurate and direct method is described for the microdetermination of a representative variety ofα-amino-acids that form soluble, partially soluble and insoluble copper-chelates of type CuA2. The method is based upon direct titration with standard copper(II) sulphate solution at a pH value ofca. 9 in presence of murexide as indicator. When compared with the indirect copper phosphate method, this direct complexometric procedure proved to be of more versatile application. Satisfactory results were obtained for 22 differentα-amino-acids with an average absolute error of ±0.1% and a maximum absolute error of ±0.3%.

Zusammenfassung

Eine direkte Methode zur Mikrobestimmung einer Reihe verschiedenerα-AminosÄuren wurde beschrieben, die lösliche, teilweise lösliche bzw. unlösliche Kupferchelate vom Typus CuA2 bilden. Die Methode beruht auf der Direkttitration mit einer CuSO4-Standardlösung bei pH∼9 in Gegenwart von Murexid als Indikator. Im Vergleich zur indirekten Kupferphos-phatmethode ist dieses komplexometrische Verfahren vielseitiger anwendbar. Für 22 verschiedene AminosÄuren wurden zufriedenstellende Resultate mit einem durchschnittlichen Absolutfehler von ±0,1% und einem maximalen Absolutfehler von ±0,3% erhalten.
