Zusammenfassung
Es wird über Untersuchungen zur Isolierung von biologisch131Jodmarkiertem Thyroxin und Trijodthyronin aus dem Blutserum mit dem Anionenaustauscher Dowex 1×2 berichtet. Diese Arbeitstechnik liefert im Vergleich zu der bisher üblichen Butanolextraktion praktisch übereinstimmende Ausbeuten in der Isolierung. Die Abtrennung der Schilddrüsenhormone von den Serumproteinen ist scharf. Eluiert wird mit einem Gemisch von konz. Ameisensäure und konz. Essigsäure. 95% der vom Austauscher gebundenen Hormone können damit rückgewonnen werden.
Die Ergebnisse führen zu einer Methodik, die im Gegensatz zur Extraktion mit Butanol geeignete Ausgangslösungen für die papierchromatographische Trennung und radioautographische Identifizierung der Schilddrüsenhormone liefert.
Summary
A report is given of studies dealing with the isolation of biological iodine-131 labeled thyroxine and triiodothyronine from blood serum with the aid of the anion exchanger Dowex 1×2. This procedure gives yields that are practically the same for the isolation as compared with the butanol extraction ordinarily used hitherto. The separation of the thyroid hormones from the serum proteins is sharp. The elution is accomplished with a mixture of concentrated formic acid and concentrated acetic acid. In this way 95% of the hormones bound by the exchange resin were recovered.
The findings lead to a method which in contrast to extraction with butanol delivers starting solutions suitable for the paper chromatographic separation and radioautographic identification of the thyroid hormones.
Résumé
La communication traite d'expériences d'isolement de la thyroxine et de la triiodothyronine marquées biologiquement par l'iode-131, à partir du sérum sanguin avec l'échangeur anionique Dowex 1×2. Cette technique, comparée à celle de l'extraction par le butanol habituellement utilisée jusqu'ici, fournit un rendement de l'isolement pratiquement identique. La séparation des hormones de la glande thyroïde d'avec les protéines du sérum est exacte. On réalise l'élution avec un mélange d'acide formique et d'acide acétique concentrés. On peut ainsi récupérer 95% des hormones fixées sur l'échangeur. Les résultat conduisent à une technique opératoire qui, contrairement à l'extraction par le butanol, fournit des solutions directement utilisables pour la séparation par chromatographie sur papier et pour l'identification radioautographique des hormones de la glande thyroïde.

J.82, 111 (1962) eine Mitteilung mit dem Titel „The Separation of Iodinated Tyrosines and Thyroxine from Serum“. Die Autoren isolieren von Serumin vitro gebundenes131J-Thyroxin mit Dowex 1×2 gefüllten Säulen. Ihre Methode führt bei Vergrößerung der eingesetzten Serummenge über ein Milliliter zu steigenden Verlusten in der Isolierung von Thyroxin. Die Ursache liegt nach Ansicht der Autoren in einer Konkurrenzierung der Bindungskräfte für Thyroxin des Austauschers mit jenen des Serumproteins.
