Zusammenfassung
Die vorliegende Arbeit beabsichtigt, eine Übersicht über die Methodik und die Anwendung der freien und der Zonenelektrophorese zu geben. Bei der freien Elektrophorese wird als Nachweismethode für die wandernden Fronten ein Verfahren vonSvensson geschildert, das gleichzeitig eine Integral- und eine Differentialkurve des Brechungsquotienten in Abhängigkeit von der Küvettenhöhe liefert. Als Beispiel für die freie Mikroelektrophorese wird die Apparatur nachAntweiler genauer besprochen. Daran schließt sich ein Überblick über die Anwendungsmethoden der freien Elektrophorese und eine Diskussion der Vor- und Nachteile dieser Methode. Bei der Zonenelektrophorese wird hauptsächlich auf die Papierelektrophorese eingegangen, wobei die Trennmöglichkeiten bei niedrigen und hohen Spannungsgefällen diskutiert werden. Als Beispiel werden hauptsächlich Trennungen von niedrigmolekularen Ionen angeführt. Schließlich werden Geräte zur quantitativen Auswertung von Elektropherogrammen auf Filtrierpapier besprochen.
Summary
The objective of the present study is to survey the methodology and the application of free and zone electrophoresis. For free electrophoresis, a description is given of a detection method for the migrating front worked out bySvensson, which yields simultaneously an integral and differential curve of the refractive quotients in relation to the height of the cell. The apparatus ofAntweiler is discussed in some detail as an example of micro electrophoresis. To this is added a survey of the application methods of free electrophoresis and a discussion of the advantages and disadvantages of this method. With respect to zone electrophoresis stress is laid principally on paper electrophoresis and the possibilities of separation at low and high potential falls are discussed. Separations of low molecular ions are cited primarily as examples. In conclusion, devices for the quantitative employment of electropherograms on filter paper are described.
Résumé
L'objet du présent travail est de donner un aperçu sur les méthodes et l'emploi de l'électrophorèse libre et de zone. Pour l'électrophorèse libre on esquisse la technique deSvensson qui permet d'identifier les fronts en migration; elle fournit simultanément une courbe intégrale et une courbe différentielle de l'indice de réfraction qui dépendent de la hauteur de la cuve. L'appareil deAntweiler est décrit avec plus de détails comme exemple de la microélectrophorèse libre. On y joint un aperçu sur les méthodes d'application de l'électrophorèse libre ainsi qu'une discussion sur leurs avantages et leurs inconvénients. En ce qui concerne l'électrophorèse de zone on insiste principalement sur l'électrophorèse sur papier au sujet de laquelle on présente une discussion relative aux possibilités de séparations sous chutes de tension faibles ou élevées. A titre d'exemple, on cite principalement les séparations d'ions de faibles molarité. On montre finalement des appareils permettant l'exploitation quantitative d'électrophérogrammes sur papier filtre.
