Zusammenfassung
Die Anwendbarkeit einiger thermodynamischer Regeln in bezug auf polymorphe Systeme wurde an 5 Modifikationen der Flufenaminsäure und an 2 Modifikationen der Mefenaminsäure geprüft. Diese Regeln setzen die Umwandlungs- oder Schmelzwärmen, IR-Spektren bzw. Dichten der Modifikationen in Beziehung zu ihrem Stabilitätsverhalten. Bei den beiden Antiphlogistika wurden daher vor allem quantitative DSC-Messungen und qualitative Löslichkeitsuntersuchungen (mit dem Thermomikroskop) durchgeführt sowie IR-Spektren aufgenommen. Die Mod. III der Flufenaminsäure ist gegenüber Mod. I und II enantiotrop; alle anderen Modifikationen dieses komplizierten polymorphen Systems sind gegeneinander monotrop bzw. ihr Umwandlungsverhalten ist ungewiß. Bei Raumtemperatur nimmt die Löslichkeit also in folgender Reihenfolge zu: III (Fp. 126° C), I (Fp. 134° C), II (Fp. 128° C), V (Fp. 122° C), VIII (Fp. 108±5° C). Die bei Raumtemperatur stabile Mod. II (Fp. 215–220° C) der Mefenaminsäure hat den Umwandlungspunkt mit Mod. I (Fp. 227–230° C) bei 130±10° C. In keinem Fall ist eine der Regeln verletzt; allerdings lieferte die „IR-Regel“ bei der Flufenaminsäure nur im Fall III/I eine signifikante Aussage.
Summary
The applicability of some thermodynamic rules referring to polymorphic systems was examined with 5 modifications of flufenamic acid and 2 modifications of mefenamic acid. These rules correlate the heats of transit'on or fusion, i. r. spectra or densities of the modifications, respectively, with their stability behaviour. Thus, the two antiphlogistics were investigated mainly by quantitative DSC and qualitative solubility determinations (by thermomicroscopy) as well as by i. r. spectroscopy. Mod. III of flufenamic acid is enantiotropic with respect to mods. I and II; all other modifications of this complicated system are monotropic to each other or their transition behaviour is not known. Thus, at ambient temperature the solubility increases in the order III (mp. 126° C), I (mp. 134° C), II (mp. 128° C), V (mp. 122° C), VIII (mp. 108±5° C). The transition point of the roomtemperature stable mod. II (mp. 215–220° C) of mefenamic acid into mod. I (mp. 227–230° C) is at 130±10° C. There is no evidence that a rule is violated in any case; for flufenamic acid, the “i. r.-rule”, however, was significant only in the case III/I.
