Zusammenfassung
Es wurde die biologische Aktivität der t-RNS nach Bestrahlung mit UV-Licht, Röntgenstrahlen und sichtbarem Licht in Gegenwart von Thiopyronin bestimmt. Dabei zeigte sich, daß bei den Aminosäuren Phenylalanin, Serin, Lysin, Leucin und Prolin durch die verschiedenen Strahlenarten die Akzeptor- und Transferaktivität etwa in dem gleichen Ausmaß zerstört werden.
Unter Berücksichtigung der spezifischen Basenzerstörung durch die drei Strahlenarten ergibt sich theoretisch eine unterschiedliche Beeinflussung der Akzeptor- und Transferaktivität, die mit der gefundenen gut übereinstimmt. Weiterhin kann man aus den Ergebnissen schließen, daß vermutlich die gleiche Basensequenz sowohl für die Aufnahme als auch Übertragung einer Aminosäure verantwortlich ist.
Summary
The biological activity of transfer RNA after its irradiation with u.v. light and X-rays, and photodynamic treatment with thiopyronine was studied. Our results as a whole indicate that a change or destruction of a base at the template site (anticode) inhibits not only the amino acid transfer activity but also the acceptor functions of t-RNA.
The specific destruction of pyrimidines caused by u.v.- and X-ray irradiations and that of guanine alone by the photodynamic action of thiopyronine show that the complimentarity between the baze sequences of m-RNA and t-RNA is unquestionable. Whether there is any correlation between the bases at the recognition — and at the template sites of t-RNA molecule is not sure but the damages at both the sites were found to be parallel.
