Zusamanfassung
Sechzehn Wirkstoffe handelsüblicher Betareceptoren-Blocker (Acebutolol, Alprenolol, Bunitrolol, Carazolol, Labetalol, Metipranolol, Metoprolol, Nado1ol, Oxprenolol, Penbutolol, Pindolol, Propranolol, Timolol und Toliprolol) wurden auf ihre Dansylierbarkeit unter-sucht.
Unter optimierten Bedingungen ist die sekundäre Aminogruppe der aliphatischen Propranolamin-Seitenkette dan-sylierbar, sofern ihr Substituent ein Isopropylrest ist. Beta-blocker, wie Bupranolol, Bunitrolol, Nadolol, Penbutolol und Timolol, die an der sekundären Aminogruppe einen tertiären Butylrest aufweisen, können mit Dansylchlorid nicht derivatisiert werden.
Die dansylierbaren Betablocker werden durch zwei-dimensionale Düinnschicht-Chromatographie vollständig getrennt und üiber ihre Rf-Werte charakterisiert. Die visuelle Nachweisgrenze liegt pro Blocker und Substanzfleck zwischen 0,2 und 10 ng/-tl. Am empfindlichsten krnnen Acebutolol, Pindolol und Carazolol nachgewiesen werden.
Summary
The suitability for dansylation was investigated of sixteen commercially available beta receptor blockers (acebutolol, alprenolol, atenolol, bunitrolol, bupranolol, carazolol, labetalol, metipranolol, metoprolol, nadolol, oxprenolol, penbutolol, pindolol, propranolol, timolol and toliprolol).
It is possible to dansylate the secondary amino group of the aliphatic propranolamine side chain under optimized conditions if it is substituted with an isopropyl residue. Beta blockers such as bupranolol, bunitrolol, nadolol, penbutolol and timolol, whose secondary amino group bears a tertiary butyl residue, cannot be derivatized with dansyl chloride.
The beta blockers that can be dansylated are completely separated by two-dimensional thin layer chromatography and are completely characterized by their Rf-values. The visual limit of determination on the silica gel plate is between 0,2 and 10 ng/gl per blocker and zone. Acebutol, pindolol and carazolol are the most sensitively detectable blockers.
