Summary
Histamine, the concentration of which may be an indicator of freshness, can be determined quantitatively by isotachophoresis (displacement electrophoresis). For sample preparation in a range greater than 40 ppm. only a simple extraction with dilute hydrochloric acid is necessary; in the range below 40 ppm, a concentration step is necessary for which we recommand freeze drying. In the concentration range of 50–100 ppm. the coefficient of variation using 5 determinations per sample is less than 2%. The rate of recovery has a mean value of 99%.
Mackerel, tuna and herring show nearly the same characteristics during storage at 4 °C. Till the third day of storage the histamin-concentration of all three samples is below 100 ppm, later the content shows a very steep increase, so that at the beginning of the 5th day the samples can be considered as spoiled. Approximately 30 min are required for each histamine determination.
Zusammenfassung
Mit Hilfe der Isotachophorese (displacement electrophoresis) ist es möglich, Histamin, dessen Gehalt als Indikator für den „Frischegrad” dienen kann, quantitativ zu erfassen. Bei Konzentrationen über 40 ppm beschränkt sich die Probenvorbereitung auf eine einfache Extraktion mit verdünnter Salzsäure. Konzentrationen unter 40 ppm erfordern einen Anreicherungsschritt, für den wir die Gefriertrocknung empfehlen. Bei den von uns durchgeführten Bestimmungen in Makrele, Thunfisch und Hering, in einem Konzentrationsbereich von 50–100 ppm, liegt der Variationskoeffizient in jedem Falle unter 2%.
Die Fischsorten Makrele, Thunfisch und Hering verhalten sich während der Lagerung bei 4 °C ähnlich. Bis zum 3. Tag liegen die Histamin-Konzentrationen bei allen Proben unter 100 ppm, danach steigt der Gehalt außerordentlich steil an, so daß am 5. Tag schon Werte erreicht werden, die die Proben als verdorben erscheinen lassen. Der zeitliche Aufwand hängt ab von der vorliegenden Histaminkonzentration und damit von der Wahl der geeigneten Trenncapillare. Er liegt in der Größenordnung von 30 min pro Bestimmung.
