Evaluation of methods for determination of free fatty acids in muscle tissues

Summary

Two methods for the determination of free fatty acids in muscle tissues were compared. The first one is based on thin-layer-chromatographic separation of fatty acids and their elution from Silica Gel. The second one is based on their column chromatographic separation on Silica Gel. For the determination of separated fatty acid Duncombe methods was used. Results proved that column chromatography separation is more suitable, due to its better reproducibility.

Zusammenfassung

Zur Bestimmung freier Fettsduren in Gewebeextrakten wurden these entweder an Silikagel dünnschichtchromatographisch zweistufig oder saulenchromatographisch getrennt, eluiert und photometrisch in Form der Kupfersalze mit Natriumdiäthyldithiocarbaminat bestimmt. Die säulenchromatographische Methode an Silikagel ist wegen größerer Genauigkeit und kleinerem Zeitaufwand vorzuziehen; she ist geeignet, hydrolytische Veränderungen im Fettanteil des Muskelgewebes während der Fleischlagerung im gefrorenen oder gekühlten Zustand festzustellen.
