Peptide Patterns of the Glutelin Fractions of Different Wheat Varieties

Summary

The glutenins of five wheat varieties with different baking quality (Kolibri, Monopol, Clement, Maris Huntsman, Durum) were partially hydrolyzed with alpha-chymotrypsin, and the soluble portions of the hydrolysates were chromatographed on Sephadex G50. The high-molecular-weight peptide fractions could be separated into a hydrophilic and a hydrophobic peptide group by high-performance liquid chromatography on octadecyl silica gel. Amount and amino-acid composition, especially the glycine content of the high-molecular-weight, hydrophilic peptide group are correlated to the baking quality of the corresponding flours.

Zusammenfassung

Die Glutelinfraktionen von fünf Weizensorten mit unterschiedlichen Backeigenschaften (Kolibri, Monopol, Clement, Maris Huntsman, Durum) wurden mit alpha-Chymotrypsin partiell hydrolysiert und die löslichen Hydrolysatanteile an Sephadex G50 chromatographiert. Durch Hochdruckflüssigkeitschromatographie der hochmolekularen Peptidfraktionen an Octadecyl-Kieselgel erfolgt eine Auftrennung in eine hydrophile und eine hydrophobe Peptidgruppe. Menge und Aminosäurezusammensetzung, insbesondere der Glycin-Gehalt der hochmolekularen, hydrophilen Peptidgruppe stehen in einer engen Beziehung zu den Backeigenschaften der entsprechenden Mehle.
