Small angle X-ray scattering studies on a dissociation Product of helix pomatia α-haemocyanin

Abstract

α-Haemocyanin of Helix pomatia was split into homogeneous fragments in an ethanolamine/HCl buffer, pH 10.2 and ionic strength 0.01. These fragments were studied by small angle X-ray scattering. The following results were obtained: radius of gyration 7.4 nm, volume 588 nm3, maximal diameter of the particle 23.9 nm, radius of the spherical subunits 2.5 nm and number of the subunits 6–9. With this information models of the particles were built and their theoretical scattering curves compared with the experimental one. Concerning molecular parameters and shape of the curve the results agreed within the limits of error with a study on the twentieths of the β-haemocyanin.
