Summary
The proteins of a purified honey solution were be separated from the low molecular weight substances by ultrafiltration and diafiltration and then concentrated. For the isolation of amylase from this protein concentrate the following methods were tested: hydrophobic interaction chromatography, ion exchange chromatography and affinity chromatography. An isolation of amylase by hydrophobic interaction chromatography and ion exchange chromatography was not possible, but attempts with affinity chromatography were successful and the isolation of the amylase resulted.
Zusammenfassung
Aus einer gereinigten Honiglösung werden die Proteine von den niedermolekularen Bestandteilen durch Ultrafiltration und Diafiltration getrennt und anschließend konzentriert. Um aus diesem Proteinkonzentrat die Amylase zu isolieren, wurden Versuche mit hydrophober Wechselwirkungschromatographie, Ionenaustauschchromatographie und Affinitätschromatographie durchgeführt.
Mit der hydrophoben Wechselwirkungschromatographie konnte eine Trennung der Amylase von Invertase, Glucoseoxidase und inerten Proteinen, nicht aber von sauren Phosphatasen erreicht werden. Die Ionenaustauschchromatographie ergab eine Aufspaltung in zwei „Amylasefraktionen”, von denen zwar eine praktisch frei von Begleitaktivitäten war, deren Rechromatographie aber zu keinen reproduzierbaren Ergebnissen führte. Dagegen waren Versuche mit der Affinitätschromatographie an α-Cyclodextrin-Sepharose erfolgreich und führten zur Reindarstellung der Honigamylase.
