Zusammenfassung
Käufliche Ribonuclease wurde durch Gelchromatographie an Sephadex G-50 superfine gereinigt und anschließend 6 Std auf 120 °C erhitzt. Das so erhaltene Produkt wurde durch Gelfiltration an Bio-Gel P-100 und Sephadex G-75 superfine in erhitzte monomere Ribonuclease, dimere Ribonuclease und zwei Oligomerfraktionen unterschiedlicher Zusammensetzung aufgetrennt. Die Stabilität der Quervernetzungen gegen dissoziierende, reduzierende und oxidierende Agentien wurde durch Gelelektrophorese nach entsprechender Behandlung gezeigt. Der Gehalt an Trinitrobenzolsulfonsäure-reaktivem Lysin weist auf eine Beteiligung von Lysinresten an der Quervernetzung hin. Die tryptische Spaltbarkeit aller Proben ist besser, als nach der Reaktivität der Lysinreste zu erwarten ist. Im Dimeren und den Oligomeren werden die Abwesenheit von Lanthionin und geringe Mengen an Lysinoalanin durch Aminosäureanalyse sowie Aspartyl-lysin und Glutamyl-lysin durch Isopeptidanalyse nach enzymatischer Totalhydrolyse nachgewiesen. Die interchenare Quervernetzung erhitzter Ribonuclease ist danach auf Lysinoalanin, Aspartyl-lysin und Glutamyl-lysin zurückzuführen (0,12, 0,30 bzw. 0,50 Mol pro Mol Dimer). Die Oligomeren zeigen entsprechende Ergebnisse. Ähnliche Ergebnisse bei erhitzter monomerer Ribonuclease, jedoch mit geringeren Isopeptidgehalten und ohne Lysinoalanin, deuten auf die gleichen Veränderungen und intrachenare Quervernetzung.
Summary
Commercial ribonuclease was purified by gel chromatography on Sephadex G-50 superfine and then heated 6 h at 120 °C. The resulting product was submitted to gel filtration on Bio-Gel P-100 followed by Sephadex G-75 superfine, yielding heated monomeric ribonuclease, dimeric ribonuclease, and two fractions of oligomeric ribonucleases differing in composition. Stability of the crosslinks against dissociating, reducing, and oxidizing reagents was proved by means of gel electrophoresis after appropriate treatment. Trinitrobenzene sulphonic acid-reactive lysine contents indicated a participation of lysine residues in crosslinking. Tryptic digestibility of all samples was better than expected with regard to lysine reactivity. Amino acid analysis demonstrated minor amounts of lysinoalanine and the absence of lanthionine, whilst isopeptide analysis after enzymic total hydrolysis demonstrated both aspartyllysine and glutamyllysine in the dimer and the oligomers. Hence intermolecular crosslinking of heated ribonuclease is attributed to lysinoalanine, aspartyllysine, and glutamyllysine (0.12, 0.30, and 0.50 moles per mole of dimer, respectively, the oligomers showing corresponding results). Heated monomeric ribonuclease exhibited similar results with smaller isopeptide contents and no lysinoalanine, indicating the same alterations and intermolecular crosslinks.
