Summary
Hydroxycinnamic acid esters with glucose or aldaric acids were determined as naturally occurring phenolic compounds in the peel and pulp of orange, grapefruit and lemon by means of analytical high performance liquid chromatography (HPLC). In the case of the hydroxycinnamic acid esters with glucose derivatives, those with ferulic and sinapic acid are dominant and those with caffeic and p-coumaric acid are minor components. Hydroxycinnamic acid esters with quinic acid, malic acid or tartaric acid were not found even at the trace level. Esters of galactaric and glucaric acid with p-coumaric or ferulic acid could be identified in orange peel. A known method for sample preparation has been applied to their extraction and separation on a polyamide column. Initially, the glucose esters were eluted quantitatively with methanol. The glucaric and galactaric acid esters were collected in a subsequent methanol/1.5% formic acid fraction. Qualitative and quantitative HPLC analyses were performed on reserve-phase material using gradient elution with 2% acetic acid in methanol. Supplementary investigations showed relationships between the identified glucaric acid esters and the additional peaks in the chromatograms. There are probably further isomeric forms. The composition of hydroxycinnamic acid derivatives in fruit and peel extracts of the threeCitrus species investigated was compared. Esters of galactaric and glucaric acid with hydroxycinnamic acids were found inCitrus fruits for the first time.
Zusammenfassung
Als natürlich vorkommende phenolische Substanzen inCitrus wurden Hydroxyzimtsäureester mit Glucose oder einer Glykarsäure in Schale und Frucht von Orange, Grapefruit und Zitrone mit analytischer HPLC nachgewiesen. Bei den Hydroxycinnamoylglucosen sind Feruloyl- und Sinapoylglucose Hauptkomponenten und Caffeoyl- und p-Cumaroylglucose Nebensubstanzen. Hydroxyzimtsäureester mit Chinasäure, Apfelsäure oder Weinsäure wurden selbst in Spuren nicht nachgewiesen. Ester von Galaktar- und Glucarsäure als Alkoholkomponenten mit p-Cumarsäure oder Ferulasäure wurden aus Orangenschalen identifiziert. Eine bekannte Methode der Probenaufarbeitung wurde für die Extraktion und Trennung über Polyamid herangezogen. Zuerst lassen sich Glucoseester quantitativ mit Methanol eluieren. In einer folgenden Fraktion mit 1,5% Ameisensäure in Methanol werden die Glykarsäureester erhalten. Die qualitative und quantitative Bestimmung mit HPLC erfolgte auf RP-18-Umkehrphasenmaterial (Gradientenprogramm) mit Methanol — 2% Essigsäure. Ergänzende Untersuchungen zeigten einen Zusammenhang zwischen den identifizierten Glucarsäureestern und zusätzlichen Substanzpeaks in den Chromatogrammen. Vermutlich handelt es sich um weitere isomere Formen. Frucht- und Schalenextrakte der drei genanntenCitrus-Arten wurden untereinander an Hand ihrer Hydroxyzimtsäureverbindungen verglichen. Glykarsäureester der Hydroxyzimtsäuren wurden inCitrus zum ersten Mal gefunden.
