The binding of imipramine, chlorimipramine, desipramine, dosulepine, amitriptyline and prothipendyl to bovine serum albumin was determined by means of sephadex gel filtration and equilibrium dialysis.

The albumin binding of the drugs was characterized by the following parameters: the free drug percentage (α), percentage of drug bound (β), the binding constants K1 k+ andm as well as the free energyΔ F°.

The partition coefficientP between n-octanol and buffer solution was determined for the above mentioned drugs as well as for promazine, triflupromazine, chlorpromazine, methopromazine, and acepromazine.

The substituent constantπ was calculated for the-CF3,-CI,-OCH3, and -OCCH3 functions.

The free drug percentageα calculated with the aid ofπ andΣπ values agreed very well with values determined experimentally except for theα values of desipramine.

It was concluded that the hydrophobic interaction of the drugs with the albumin plays the main role, but evidence for steric effects and ionogenic binding was also found.
