Gonadotrophins and their estimation

Summary

A short survey of the chemical and physiological properties of gonadotrophins is given, and some problems in relation to the biological estimation of gonadotrophins are commented upon. The general criteria for good assay methods, such as specificity, sensitivity, accuracy, simplicity are described. Methods used or advocated for the separate estimation of various gonadotrophic principles, such as follicle stimulating hormone (FSH), interstitial cell stimulating hormone (ICSH), luteotrophic hormone (LTH), human menopause gonadotrophin (HMG), human chorionic gonadotrophin (HCG) and pregnant mare's serum gonadotrophin (PMS) are critically discussed and recommendations are made on methods, which are considered to be suitable for clinical assays of urinary gonadotrophins.

A discussion of some of the variables affecting bioassays of gonadotrophic hormones, such as species and strain differences, age and weight of animals, caging, diet, route of administration, schedule of dosage, time of sacrifice, seasonal variations etc. is also included. Methods suggested for “chemical” estimation of gonadotrophins are also reviewed and critically discussed. Published methods for the extraction, concentration and separation of urinary gonadotrophins are tabulated and a detailed technical description of an extraction technique recently developed byLoraine andBrown (1956) is also presented. The last chapter reviews the clinical conditions, in which the estimation of urinary gonadotrophins may be of diagnostic value.
