RNA synthesis during morphogenesis of the fungusMucor racemosus

Abstract

Individual rates of synthesis were determined for the major classes of RNA throughout the yeast-tohyphae transition ofMucor racemosus. Cells at stages throughout development were pulse-labelled with3H-uracil or32P-phosphoric acid, broken under liquid nitrogen and extracted with phenol to isolate the RNA. The major classes of RNA (tRNA, rRNA, precursor rRNA) were separated and quantified on sucrose density gradients. Polyadenylated RNA was identified and quantitated after passage through an oligodeoxythymidylic acid-cellulose column. All species of RNA showed an increased rate of synthesis immediately after the atmospheric shift (CO2-to-air) used to initiate development. All rates accelerated until maximums were observed at the time of hyphal germ tube emergence. Thereafter, during hyphal elongation, the rates decelerated. During the period of most rapid synthesis the ribosomal forms of RNA showed a proportionally higher increase in rate of synthesis than the other forms. Newly synthesized mRNA remained a constant fraction of the total RNA made at all time throughout the morphogenesis.
