Zusammenfassung
Aus Retikulin, das aus normalen menschlichen Milzen isoliert wurde, wurde mit Papain ein Mucoproteid freigesetzt, das 8,4% N-Acetyl-Neuraminsäure, 4% Galaktose, 2,5% Mannose, 2,5% Fucose, 12,4% N-Acetylglucosamin, 12,1% Glucosamin und 44,6% Eiweiß enthielt. Als Aminosäuren fanden sich in größerer Menge Glutamin- und Asparaginsäure, Serin und Threonin, Glycin, Alanin, Valin, Leucin und Isoleucin. Daneben konnten Cysteinsäure, Cystin und Methionin, Arginin, Histidin und Lysin, Tyrosin und Phenylalanin, Prolin und Hydroxyprolin sowie α-Aminobuttersäure nachgewiesen werden. Weitere Mucoproteide konnten im Retikulin nicht gefunden werden.
Nach Freisetzen der Neuraminsäure sowie nach Extraktion des neuramin-säurehaltigen Mucoproteids mit Papain kommt es im Feingewebsschnitt der Milz zu einem gleich starken Abfall der Basophilie des Reticulums wie seiner Fähigkeit zur Metachromasie.
Bei Berücksichtigung der bisher vorliegenden Untersuchungsergebnisse ergibt sich für den Aufbau der Reticulumfasern folgendes Bild: Die Reticulumfaser besteht aus dünnen Kollagenfibrillen, die durch ein neuraminsäurehaltiges, ein sog. neutrales Mucoproteid miteinander verkittet sind. Außerdem enthält sie Fettsäuren, vorwiegend Myristinsäure, die gerichtet in die Faser eingelagert sind.
Summary
A mucoprotein was extracted with papain from the reticulum which was isolated from the normal human spleen. This mucoprotein contained 8.4% N-acetyl-neuraminic acid, 4% galactose, 2.5% mannose, 2.5% fucose, 12.4% N-acetylglucosamine, 12.1% glucosamine, and 44.6% protein. The more abundant amino acids found were glutamine, asparagine, serine, threonine, glycine, alanine, valine, leucine, and isoleucine. In addition, cysteine, cystine, methionine, arginine, histidine, lysine, tyrosine, phenylalanine, proline, hydroxyproline, and α-aminobutyric acid could be demonstrated. Other mucoproteins could not be found in the reticulum.
After the release of the neuraminic acid and the extraction of the neuraminic acid containing mucoprotein with the papain, there was a concomitant marked decrease in the basophilia of the reticulum and in its ability to show metachromasia in thin sections of the spleen.
In consideration of the experimental results presented, the following may be said for the composition of the reticulum fiber: it consists of thin fibrils of collagen which are cemented together with a so-called neutral mucoprotein containing neuraminic acid. In addition, it contains fatty acids, chiefly myristinic acid, which are orderly bound within the fibers.
