Zusammenfassung
Die durch enzymatische Hydrolyse von N-Acylaminosäuren freiwerdenden Aminosäuren können, ausgehend von pH 7,5, durch direkte Titration auf einen vorgewählten Endpunkt mittels einer automatischen Titrationsanlage mit hinreichender Genauigkeit quantitativ bestimmt werden. Als geeigneter Titrationsendpunkt (Äquivalenzpunkt) hat sich bei den meisten Aminosäuren pH 11,0, bei Lysin 11,50, Cystein 11,75 und Prolin 11,80 bewährt. Die Anwendung der Methode wird an praktischen Beispielen der enzymatischen Hydrolyse von N-Acylaminosäuren demonstriert.
Abstract
Amino acids enzymatically liberated from N-acylamino acids can be assayed with adequate precision by an automatic potentiometric titration from pH 7.5 to a fixed end point. For the majority of amino acids pH 11.0 proved to be the most suitable end point, whereas the respective value for leucine was pH 11.50, for cysteine pH 11.75 and for proline 11.80. Two representative examples are given for the applicability of the method.
