Summary
The development of various species of Echinostomatidae in snails of the genusLymnaea was investigated by histological methods. Attention was focussed on the differences between the first infection and the homologous reinfection or the heterologous superinfection, which reveal intensely decreasing infection rates. During reinfection the penetration of miracidia decreased to 35%, the amount of miracidia encapsulated by fibrocytes instead of amoebocytes rose by 73% and 33% of them were destroyed. Superinfection was equally difficult:Lymnaea showed resistance againstIsthmiophora melis if the first infection had been performed with another species of Echinostomatidae, even if the first infection did not develop. The results indicate that the reaction is rather non-specific.
Unlike other host-parasite relationships this acquired resistance is a pure cellular reaction without any dependence upon humoral factors. Survival of miracidia ofIsthmiophora melis incubated in haemolymph of infected and uninfected hosts were identical, whereas haemolymph ofBiomphalaria glabrata killed the miracidia within a few seconds. The dependence of the resistance upon the amount of blood cells was investigated by bleeding or irradiating the snails by X-rays. None of these treatments increased the sensibility of the snails against miracidia. The non-specifity of this acquired resistance was assayed by injecting some foreign substances into the snail's head-foot. These non-specific substances did increase the resistance almost as much as an already existing echinostomatid-infection.
Zusammenfassung
Um das Problem der im Vergleich zur Erstinfektion sowohl bei (homologer) Reinfektion wie bei (heterologer) Superinfektion auf fast 1/3 absinkenden Infektionsrate zu lösen, wurde mit histologischen Methoden die Entwicklung mehrerer Enchinostomatidenarten in lymnaeiden Schnecken verfolgt.
Es zeigte sich, daß bei der Reinfektion vonLymnaea stagnalis mitIsthmiophora melis nur noch etwa 1/3 der verwendeten Miracidien penetriert, daß die Zahl der Miracidien, die von Fibrocyten statt von Amoebocyten eingekapselt wird, um 73% ansteigt und daß die wenigen penetrierten Miracidien zu mehr als 30% abgebaut werden.
Im gleichen Ausmaß wird auch die Superinfektion erschwert, was auf eine gewisse Unspezifität dieser Reaktion hinweist:Lymnaea erwirbt Resistenz gegenIsthmiophora auch dann, wenn die Erstinfektion mit einer anderen Art derselben Trematodenfamilie gesetzt wurde, selbst wenn diese nicht angegangen ist.
Im Gegensatz zu anderen Parasit-Wirt-Verhältnissen ist diese erworbene Resistenz als rein zelluläre Reaktion nicht von humoralen Faktoren abhängig, denn Miracidien überlebten in Hämolymphe von infizierten Wirten genau so lange wie in Hämolymphe von uninfizierten Tieren. Zum Vergleich diente in diesem Versuch Hämolymphe vonBiomphalaria glabrata, die die Miracidien in Sekundenschnelle abtötet.
Da die phagocytierenden Blutzellen für die erworbene Resistenz verantwortlich sind, wurden sie durch Blutverlust oder Röntgenbestrahlung ausgeschaltet. Derartige Vorbehandlungen vor der Infektion konnten aber die Infizierbarkeit vonLymnaea stagnalis nicht steigern.
Die Unspezifität der erworbenen Resistenz wurde geprüft durch Injektion verschiedener Fremdstoffe. Auch diese völlig unspezifischen Reize hatten aufLymnaea stagnalis ähnliche resistenzsteigernde Auswirkungen wie eine bereits bestehende Echinostomatideninfektion.
