Zusammenfassung
N-Acetyl-d-neuraminsäure wurde mit Acetanhydrid acetyliert, die erhaltene 2,4,7,8,9-Penta-O-acetyl-N-acetyl-d-neuraminsäure mit trockenem Chlorwasserstoff in die 4,7,8,9-Tetra-O-acetyl-N-acetyl-2-chlor-2-deoxy-d-neuraminsäure übergeführt und diese mit verschiedenen aliphatischen, araliphatischen und hydroaromatischen Alkoholen in Gegenwart säurebindender Mittel umgesetzt; schließlich wurden die O-Acetylgruppen mit verdünntem Alkali entfernt. Auf diese Weise wurde eine Anzahl verschiedener Ketoside der N-Acetyl-d-neuraminsäure gewonnen, von denen nur eines, das m-Nitrobenzyl-glykosid, kristallisierte, alle jedoch gut kristallisierende Tetra-O-acetyl-Derivate gaben.
Den in dieser Arbeit beschriebenen synthetischen Ketosiden, aus denen der glykosidisch gebundene Alkohol durchVibriocholerae-Neuraminidase enzymatisch abgespalten werden kann, wird die α-Konfiguration zugeschrieben.
Abstract
N-Acetyl-d-neuraminic acid was acetylated with acetic anhydride and the resulting 2,4,7,8,9-penta-O-acetyl-N-acetyl-d-neuraminic acid treated with dry hydrogen chloride to give 4,7,8,9-tetra-O-acetyl-N-acetyl-2-chloro-2-deoxy-d-neuraminic 4,7,8,9-tetra-O-acetyl-N-acetyl-2-chloro-2-deoxy-d-neuraminic acid. The latter was reacted with various aliphatic, araliphatic and hydroaromatic alcohols, in the presence of acid-binding agents, and finally the O-acetyl groups were removed with the help of dilute allsali. Thereby a number of different ketosides of N-acetyl-d-neuraminic acid were obtained. Only one of them,viz. the m-nitrobenzyl glycoside, cristallized as such. The others, however, gave well crystallizing tetra-O-acetyl derivatives. The synthetic ketosides of neuraminic acid described in this paper, all of which are cleaved by the receptor-destroying enzyme ofVibrio cholerae with release of the glycosidically bound alcohol, are considered to be the α-anomers.
