δ-Aminolävulinsäure-Synthetase in Bäckerhefe. Vergleichende Untersuchung des Enzyms in Wildstamm und Petite-Mutante

Abstract

The activity of δ-aminolevulinic acid synthetase was determined using an assay, which allows low levels of the enzyme to be measured. Glucose repression has been excluded as a possible cause for the low values of enzyme activity found.

The activities of δ-aminolevulinic acid synthetase in the aerobically grown wild type and the respiration-deficient petite mutant derived therefrom were rather similar.

In the wild type cells as well as in those of the petite mutant only about 5% of the total δ-aminolevulinic acid synthetase activity was shown to reside in the mitochondrial fraction, the predominant portion of the enzyme being in the soluble fraction.

Zusammenfassung

δ-Aminolävulinsäure-Synthetase wurde inSaccharomyces cerevisiae mit Hilfe eines für die Messung sehr niedriger Enzymaktivitäten geeigneten Verfahrens bestimmt.

Repression durch Glucose konnte als mögliche Ursache der gefundenen niedrigen Aktivitätswerte ausgeschlossen werden.

In dem aerob gezüchteten Hefe-Wildstamm und einer aus ihm gewonnenen atmungsdefizienten Petite-Mutante war die Enzymaktivität annähernd gleich.

Sowohl in den Zellen des Wildtyps als auch in denen der Petite-Mutante wurden nur etwa 5% der gesamten zellulären Enzymaktivität in der Mitochondrien-, der überwiegende Teil in der löslichen Fraktion gefunden.
