Zusammenfassung
Aus dem MischpräparatLHPO wurden zwei neue Lipidhydroperoxide, Substanz 64 und 65, isoliert und als Hydroperoxycaprylsäuremethylester und Hydroperoxy-trans-nonen-(2,3)-al-(1) identifiziert. Beide Substanzen zeigen deutliche Hemmwirkungen auf Gärung und Atmung von Ehrlich-Ascites-Tumorzellen in vitro, insbesondere Subst. 65, offenbar infolge der im selben Molekül gleichzeitig anwesenden Hydroperoxy- und α,β-Enalgruppe. Der Reaktionsmechanismus von Substanz 64 in bezug auf die Gärungshemmung besteht im wesentlichen in einer Selektiv-Hemmung vonGAPDH undLDH, während dasNAD-System nicht essentiell betroffen wird.
In bezug auf die Hemmwirkungen des MischpräparatesLHPO ergibt sich, daß dessen gärungshemmende Wirkung im wesentlichen der Subst. 64 zugeschrieben werden kann, während die Atmungshemmung nicht als reiner Persauerstoffeffekt zu erklären ist. Hier wird vielmehr eine synergistische Wirkung gewisser im PräparatLHPO anwesender Carbonylverbindungen anzunehmen sein.
Abstract
Two more lipid hydroperoxides, the substances 64 and 65, were isolated from the crude preparationLHPO and were identified as methyl hydroperoxycaprylate and hydroperoxy-trans-nonen(2,3)-al-(1). Either compound is markedly active in Vitro in inhibiting both fermentation and respiration of Ehrlich ascites tumour cells, particularly substance 65 which is characterized by the simultaneous presence within the same molecule of an hydroperoxy and an α,β-enal group. The interference of substance 64 with fermentation is due essentially to a selective inhibition of glyceraldehyde phosphate dehydrogenase and lactic dehydrogenase, whereas the NAD system does not appear to be fundamentally involved.
The inhibition of fermentation by the crude preparationLHPO may be largely ascribed to substance 64, whereas the respiratory inhibition can not be accounted for by peroxy compounds exclusively, but rather by a synergistic action of several carbonyl compounds present inLHPO.
