Summary
To investigate the DNA-replication of T2 phage in singly infected cells the thymine requiring mutantE. coli B-3 was used as a host. The experiments were done to decide the question whether damages in gene functions suffice to explain the phage killing action of UV. If an additional type of damage exists directly inhibiting replication the synthesis of phage DNA should be blocked even in cells where the early phage functions can be carried out. Under the assumption that the target size of early functions is less than 60 percent of that of phage survival, the reported results provide evidence for the existence of damages directly inhibiting replication.
Single infection ofE. coli B-3 with an UV-irradiated phage does not immediately kill the cell. The phage gene controlling thymidylate synthetase allows the synthesis of bacterial DNA for a certain time. This observation is discussed in connection with the metabolic changes in the bacterium after phage infection.
