Zusammenfassung
1. Die Calciumaufnahme und -abgabe wurde an Menschen-Erythrocyten untersucht.
2. der Ca-Influx beträgt bei 37° C in Tyrodelösung 2–3 · 10−3 μÄqu/ml Erythrocyten in 1 Std. er ist der Außenkonzentration direkt proportional und nimmt mit steigender Wasserstoffionenkonzentration zu.
3. Bei 3° C ist die Durchlässigkeit der Erythrocytenmembran für Ca erhöht. Der Ca-Gehalt der Erythrocyten steigt während einer 24 stündigen Inkubation in isotoner CaCl2-Lösung auf 8 μÄqu/ml Erythrocyten an. Gleichzeitig nimmt der K-Gehalt der Erythrocyten von 94,5 auf 45–50 Äqu/ml, d. h. um trund 50% ab. Die Ca-Aufnahme in der Kälte ist von der Ca-Außenkonzentration abhängig, jedoch besteht keine lineare Beziehung. erst bei Ca-Außenkonzentrationen von mehr als 30 μÄqu/ml kommt es zu einem Anstieg des Ca-Gehaltes der Erythrocyten. Bei höheren Konzentrationen nimmt die Ca-Aufnahme steil zu.
4. Die Ca-Abgabe von Erythrocyten, die bei 3° C in isotoner CaCl2-Lösung vorinkubiert waren, beträgt bei 37° C 3–5 μÄqu/ml in 1 Std, das sind rund 50% des vorher in der Kälte aufgenommenen Calciums (8 μÄqu/ml Erythrocyten). Sie ist proportional der Ca-Konzentration in den Erythrocyten und ist bei 37° C und 27° C praktisch gleich.
5. Die Ca-Abgabe zeigt eine erste, schnelle Phase, während der etwa 50% des aufgenommenen Ca abgegeben werden, und eine zweite, sehr langsame Phase.
6. Strophanthin (1,36 · 10−5 m) hat keinen Einfluß auf die Abgabe des vorher bei 3° C in den Erythrocyten aufgenommenen Calciums.
Summary
Movement of Ca ions across the cell membrane was studied in human erythrocytes.
At 37° C, Ca uptake in Tyrode solution amounted to 2–3 · 10−3 μEqu/ml red cells/hour. It was directly proportional to the external Ca concentration and increased with decreasingpH. At 3° C, the permeability of the erythrocyte membrane for Ca was higher. The Ca content rose during 24 hours in isotonic CaCl2 solution to 8 μEqu/ml erythrocytes. During the same time the K content decreased by about 50%. The Ca uptake in the cold depended upon the Ca content of the medium, but the relationship was not linear. There was no change in Ca content at concentrations up to 30 μEqu/ml, with higher concentrations the uptake rose steeply.
Erythrocytes, which had been preincubated at 3° C in isotonic CaCl2 solution, lost in one hour at 37° C 3–5 μEqu/ml, i.e. about 50% of the Ca taken up in the cold. This efflux was proportional to the Ca concentration in the erythrocytes and was the same at 37° C and 27° C. Ouabain did not influence the Ca loss of preincubated erythrocytes.
