Zusammenfassung
Mit zytochemischer Methodik wird an Blutausstrichen der Einfluß verschiedener Stoffe auf die alkalische Leukozytenphosphatase (a. L.-Ph.) untersucht. Der Phosphatasenachweis wird teils mit dem Ca-Co-Verfahren, teils mit simultaner Azo-Kupplung geführt und die Aktivität des Ferments nach einem Zählmodus geschätzt.
Sicher inaktivierten Cyanid, Phosphorwolframsäure, Citrat und Ascorbinsäure, während die Wirkung von tertiärem Phosphat bei Verdünnung unter 10−2 m allmählich in eine Aktivierung umschlug. Eine wesentliche Fluoridhemmung war erst bei 10−1 m Konzentration zu erzielen.
Die erprobten Aminosäuren zeigen meist erst in einer Konzentration unter 10−3 m eine Aktivierung der a. L.-Ph. Histamin hemmte nur in relativ hoher Konzentration; Heparin, ACTH und Prednisolon zeigten keine Hemmung, eventuell eine geringe konzentrationsabhängige Aktivierung.
Methodische Einzelheiten und Rückschlüsse werden besprochen.
Summary
The effect of various compounds upon the activity of alkaline phosphatase in human leucocytes is examined by cytochemical methods. The demonstration of the sites of enzyme activity is accomplished by a modification of the Ca-Co-method and by simultaneous azo-coupling-technic. The activity of alkaline phosphatase in blood smears is estimated by a scoring technic.
Under the conditions of the experiment, the enzyme was strongly inhibited by cyanide, phosphotungstic acid, citrate, and ascorbic acid, while the inhibiting effect of tertiary phosphate gradually changed to slight activation at concentrations below 10−2 m. Fluoride was essentially inhibiting the enzyme only above concentrations of 10−2 m.
The amino-acids tested showed a certain activating effect only in concentrations below 10−3 m. Histamine inhibited the phosphatase but in relative high concentrations. Heparine, ACTH, and prednisolone did not inhibit the enzyme but were slightly activating in dependence on concentration.
