Zusammenfassung
Homogenate von Leber, Herz, Niere, Milz, Gehirn und Knochenmark sowie Suspensionen von reticulocytenreichen Blutzellen (Kaninchen) sind zur biologischen Chelation von Hämin aus Protoporphyrin und Eisen befähigt. Das entstandene Hämin ist identisch mit authentischem Hämin. Die Aktivitäts-pH-Kurve des Enzyms (Iron-Protoporphyrin-Chelating-Enzyme (IPCE)) weist zwei pH-Optima auf (pH 7,8 und 8,8). Reduziertes Protoporphyrin (Protoporphyrinogen) wird gegenüber Protoporphyrin vermindert eingebaut. Die Ermittlung der spezifischen Organaktivitäten/ Milligramm Organ(Homogenat-)Trockengewicht ergibt den größten Umsatz im Herzmuskel. Es folgen Niere, Gehirn, Leber, Milz und Knochenmark. Sowohl zweials auch dreiwertiges Eisen sind zur Chelation befähigt, wobei das dreiwertige Eisen höhere Einbauraten aufweist. Die Reaktion verläuft über mehrere Stunden, kann durch Sauerstoff gehemmt und durch Kohlendioxyd und (oder) reduziertes Glutathion aktiviert werden.
