Isolation and characterization of lipophorin from the hemolymph of diapausing larvae of the southwestern corn borer,Diatraea grandiosella

Summary

  1. 1.

    Lipophorin was isolated from the hemolymph of diapausing larvae of the southwestern corn borer,Diatraea grandiosella, and shown to have a density of 1:11 g/ml and a molecular weight of 520,000.

  2. 2.

    The holoprotein was comprised of apolipoprotein-I (M r=231,000) and apolipoprotein-II (M r=74,000), and contained about 38% lipid (28% neutral lipids, 10% polar lipids), and 3% carbohydrate.

  3. 3.

    The major neutral lipids present were diacylglycerol and sterol, while the major polar lipids present were phosphatidyl choline, phosphatidyl ethanolamine, and sphingomyelin. Carotenoids were also present.

  4. 4.

    Apolipophorin-I and apolipophorin-II were immunochemically distinct. The holoprotein was immunochemically identical to a protein present in the larval hemolymph of two other species ofDiatraea.

  5. 5.

    A mean concentration of 5 mg lipophorin/ml hemolymph was determined using rocket immunoelectrophoresis.
