Summary
Pooled maternal-, retroplacental- and cord serum were purified by means of fractionation with Rivanol and ammonium sulfate, simple and recyclic Sephadex G-200 gel filtration, DEAE-Sephadex A-25 and A-50 chromatography and electrofocusing with Ampholine pH 3–10 and pH 4–6. The pregnancy-typical proteins in these fractions were demonstrated with a mixture of absorbed rabbit antisera raised against maternal serum and placental tissue extract by means of immunodiffusion (Ouchterlony) and immunoelectrophoresis (Scheidegger). Fractionation with ammonium sulfate in connection with recyclic Sephadex G-200 gel filtration was an effective procedure. Some protein fractions immunoelectrophoretically contained one pregnancy-typical protein and only two normal serum proteins. Molecular weights of two pregnancy-typical proteins could be estimated approximately.
Zusammenfassung
Zur Isolierung schwangerschaftstypischer Proteine wurden gepooltes Sehwangeren-, Retroplacentar- und Nabelschnurserum unter Anwendung der Salzfällung mit Rivanol und Ammoniumsulfat, der einfachen und recyclischen Gelfiltration mit Sephadex G-200, der Ionenaustauschchromatographie an DEAE-Sephadex A-25 und A-50 sowie der Elektrofokussion mit Ampholine pH 3–10 und pH 4–6 fraktioniert. Die in den Fraktionen enthaltenen schwangerschaftstypischen Proteine wurden mit einer Mischung absorbierter Kaninchenantisera gegen Schwangerenserum und Placentaextrakt in der Immunodiffusion nach Ouchterlony und der Immunelektrophorese nach Scheidegger nachgewiesen. Als effektives Verfahren erwies sich die Salzfraktionierung mit Ammoniumsulfat in Verbindung mit der recyclischen Gelchromatographie. Es konnten Proteinfraktionen gewonnen werden, die neben einem schwangerschaftstypischen Protein immunelektrophoretisch nur zwei Normalserumproteine enthalten. Aus dem Verhalten bei der Fraktionierung konnten Rückschlüsse auf das Molekulargewicht der schwangerschaftstypischen Proteine gezogen werden.
