Zur Wirkung von LH auf den Stoffwechsel isoliert perfundierter menschlicher Ovarien

The effect of LH on the metabolism of isolated perfused human ovaries

Summary

Eight human ovaries with graafian follicles, one of them with a diabetes mellitus, were perfused, in vitro, with a 100% synthetic hemoglobin-free medium and stimulated with LH. The oxygen uptake of the ovaries and the changes in the level of glucose, pyruvate and lactate in the perfusion medium during its five hour circulation were determined. Additionally we tested the activities of the GOT and GPT. As criteria of viability of the perfused ovaries the concentrations of ATP, ADP, glucose, pyruvate, lactate, glycogen and the activities of the GOT, GPT and G-6-PDH were determined in the tissue after the close of the perfusion. The stimulation with 1 μg LH-NIH-S12/ml pm caused an increase in glucose-assimilation and lactate-production, but a decrease in pyruvate-consumption and only a small rise in oxygen uptake. The metabolism of the ovary seems to be interfered in diabetes mellitus. The results confirm the possibility to maintain an intact metabolism in the ovaries for a perfusion-period of five hours.

Zusammenfassung

Acht menschliche Ovarien mit Graafschem Follikel, eines davon bei schwerem Diabetes mellitus, wurden in einem geschlossenen, rezirku-lierenden System mit einem vollsynthetischen, hämoglobinfreien Medium isoliert in vitro über 5 Std perfundiert und mit LH stimuliert. Während der Perfusion wurde der O2-Verbrauch ermittelt, sowie im Medium die Konzentrationsänderungen von Glucose, Pyruvat und Lactat. Zusätzlich wurden die Aktivitäten der GOT und GPT gemessen. Als Kriterium der Intaktheit des Gewebes nach Perfusionsschluß bestimmten wir im Ovar den Gehalt an ATP, ADP, Glucose, Pyruvat, Lactat und Glykogen sowie die Aktivitäten der GOT, GPT und G-6-PDH. Es konnte gezeigt werden, daß die Ovarien auf eine LH-Stimulation mit einer Zunahme der Lactat-produktion und Glucoseaufnahme trotz aerober Bedingungen reagierten und die Pyruvataufnahme dabei gleichzeitig herabgesetzt war. Bei Steigerung der LH-Dosis um das 4fache nahmen Lactatproduktion und Glucoseaufnahme beide um einen Faktor 1,2 zu, während Pyruvataufnahme und O2-Verbrauch eine nicht so deutliche Änderung zeigten. Bei diabetischer Stoffwechsellage scheint der Ovarmetabolismus beeinträchtigt zu sein. Die Ergebnisse bestätigen die Möglichkeit, in isoliert perfundierten Ovarien über 5 Std einen zu spezifischen Leistungen noch befähigten Stoffwechsel aufrechtzuerhalten.
