Summary
The esterase activity is determined by lowering the pH-value. One prerequisite is a suitable indicator system which guarantees a linear relationship between extinction and pH-value in the specified pH-range. Further, the esterase should not have too high a pH-dependency under reaction conditions. The indicator used was bromochlorophenolblue. The measuring scale was from pH 4.5 to 3.9. Analysis was performed in the stop-flow mode on the Tecator FIAstar 5020 analyzer, obtaining a sample frequency of 15 samples per hour. The variation coefficient obtained for this process was 4.5%.
