Enzyme cytochemical studies on cell suspensions and primary cultures of guinea pig epidermis

Summary

Epidermal cell suspensions and primary cultures from adult guinea pigs were subjected to a variety of enzyme cytochemical reactions. The suspensions were regularly found to contain, apart from keratinocytes in their various stages of differentiation, Langerhans cells and a number of fibroblasts. These latter cell types were observed up to the third day of culture but were apparently unable to survive under routine conditions of cell culture. The enzyme cytochemical profile of the heavily proliferating cell type proved to be identical with that of the basal layers of the epidermis in vivo.

Zusammenfassung

Zellsuspensionen und primäre Zellkulturen aus der Epidermis des erwachsenen Meerschweinchens wurden einem ausgewählten Spektrum enzymcytochemischer Reaktionen unterworfen. Es wurde festgestellt, daß neben den verschiedenen Differenzierungsstufen der Keratinocyten auch stets Langerhans-Zellen und einige Fibroblasten in der Suspension enthalten sind, bis zum dritten Tag der Kultur nachgewiesen werden können, aber offensichtlich unter Routinebedingungen nicht überleben. Die Kulturzellen besitzen dasselbe enzymcytochemische Profil wie die Keratinocyten der basalen Zellagen in vivo.
