The effects of parenteral injections of glucocorticosteroids on Guinea pig epidermis pretreated with Vitamin A-Acid (VAA) have been investigated autoradiographically (incorporation of3H-Thymidin into DNA) and biochemically (measurement on the DNA-dependent RNA-polymerase activity of the epidermis).

Pretreatment of the animals with glucocorticosteroids prevents the development of VAA-induced abnormalities in keratinization.

The stimulation of epidermal RNA synthesis by VAA was further increased by low concentrations of glucocorticosteroids. Higher concentrations however, inhibited RNA synthesis as indicated by the activity of the DNA-dependent RNA-polymerase.

The effects of glucocorticosteroids on epidermal RNA synthesis are rapid and dose-dependent.

Mit Hilfe autoradiographischer (3H-Thymidin Markierung der DNS) und biochemischer (Messung der DNS-abhängigen RNS-Polymerase-Aktivität der Epidermis) Analysenverfahren wurde die Wirkung von parenteral verabfolgten Glucocorticoiden auf die Vitamin A-Säure (VAS)-vorbehandelte Meerschweinchenepidermis untersucht.

Nach Vorbehandlung der Versuchstiere mit Glucocorticosteroiden konnten die charakteristischen strukturellen Merkmale einer gestörten Verhornung nicht beobachtet werden.

Die Steigerung der VAS-bedingten epidermalen RNS-Synthese wurde durch niedrige Glucocorticoid-Konzentrationen verstärkt. Dagegen bewirken höhere Hormondosen eine Hemmung der RNS-Synthese gemessen an der Aktivität der DNS-abhängigen RNS-Polymerase.

Es handelt sich um frühzeitige und dosisabhängige Beeinflussung der epidermalen RNS-Synthese durch Glucocorticosteroide.
