The bile ducts of rats were occluded for up to 18 days. Over this period of investigation the maximum pressure attainable in the bile duct fell from approximately 21 to 10 cm H2O after 4 to 6 days. During cholestasis these maximum pressure values were continually maintained. After cholestasis the bile flow was already considerably restricted against a moderately increased external pressure. This observation as well as regurgitation studies in rats previously subjected to cholestasis, indicate that the fall of the maximum bile pressure is caused by a reduction of the regurgitation resistance. From these results we may conclude that during chronic cholestasis, bile is also continuously secreted into the bile ducts but constantly flows back into the blood and into the lymph.

This result was supported by the histological observation that i. v. injected primulin is secreted into the bile capillaries in rats previously subjected to cholestasis, up to the 5th cholestatic day.

In rats whose bile ducts were occluded for up to 18 days, the bilirubin concentration was measured in the blood, in the bile contained in the biliary tract and also in the bile which left the bile duct after opening the occlusion. The results obtained indicate that there is a reversal in the direction of secretion for bilirubin in the liver cells beginning from the 2nd–4th day of cholestasis. The restoration of the original polarity of the cells takes longer as the duration of cholestasis increases.

An Ratten, deren Gallengang bis zu 18 Tagen verschlossen war, nimmt der in den Gallenwegen maximal erreichbare Druck nach 4-6 Tagen von ca. 21 auf ca. 10 cm H2O ab und bleibt für die restliche Untersuchungszeit auf gleicher Höhe. Diese Höchstwerte werden während der Gallenstauung kontinuierlich aufrechterhalten. Durch die Beobachtung, daß nach einer Gallenstauung der Gallenfluß bereits gegen mäßig erhöhten äußeren Druck erheblich eingeschränkt wird sowie durch Rückflußversuche an zuvor gallegestauten Tieren, konnte gezeigt werden, daß die Abnahme des maximalen Gallendruckes auf eine Verminderung des Rückflußwiderstandes zurückzuführen ist. Aus diesen Ergebnissen ist zu schließen, daß auch während einer chronischen Gallenstauung fortlaufend Galle in die Gallenwege sezerniert wird; sie fließt jedoch beständig in das Blut und in die Lymphe zurück.

Dieses Ergebnis wird gestützt durch die histologische Beobachtung, daß bei Ratten mit bestehender Gallenstauung bis zum 5. Stauungstag i.v. injiziertes Primulin in die Gallencapillaren ausgeschieden wird.

An Ratten, deren Gallengang bis zu 18 Tagen verschlossen war, wurde die Bilirubinkonzentration im Blut, in der in den Gallenwegen enthaltenen Galle sowie in frei abfließender Galle nach Öffnen des Verschlusses verfolgt. Die hierbei erhaltenen Ergebnisse weisen darauf hin, daß es nach 2-4 Tage währender Gallenstauung zu einer Umkehr in der Sekretionsrichtung innerhalb der Leberzellen für Bilirubin kommt. Die Wiederherstellung der ursprünglichen Polarität der Zellen nimmt nach längerer Gallenstauung mehr Zeit in Anspruch.
