Juxtaglomerular granules in subcellular fractions of guinea-pig kidney

Summary

The preparation of a subcellular fraction of renin-containing juxtaglomerular granules of guinea-pig kidney is described. Two separate preparations had a 8.4 and 15 times higher pressor activity [3] per mg protein than the respective homogenate. Electron microscopy showed mitochondria, non-specific and specific juxtaglomerular granules in the granular fraction. It is concluded, that the concept of adult guinea-pig kidney as an agranular model for the investigation of production and release of renin has to be modified.

Zusammenfassung

Es wird eine Methode zur Gewinnung angereicherter Fraktionen juxtaglomerulärer Granula der Meerschweinchenniere beschrieben. Die Endfraktion zweier Präparationen hatte, auf den jeweiligen Eiweißgehalt bezogen, bei der Bestimmung nach Boucher [3], eine 8,7 bzw. 15 mal höhere pressorische Aktivität als das Ausgangshomogenat. Die elektronenmikroskopische Untersuchung zeigte neben Mitochondrien und unspezifischen auch spezifische Renin-haltige Granula. Aus diesen Befunden ergibt sich, daß die Nieren erwachsener Meerschweinchen entgegen früheren Annahmen kein besonderes granulafreies Modell zum Studium der Synthese und Freisetzung von Renin sind.
