The assay of ACTH and the action of ACTH and steroids on corticosteroidogenesis in the mouse

Summary

The optimum conditions are described for the assay of ACTH by its steroidogenic action on mouse adrenal glands. The index of precision averaged 0.10 and the assay can detect 1 imu ACTH/ml. High concentrations of β-oestradiol, testosterone, and corticosterone inhibited adrenal steroidogenesis acting at a site prior to the formation of pregnenolone. ACTH appeared to stimulate corticosteroidogenesis between cholesterol and pregnenolone. High concentrations of 3′5′ AMP also stimulated the glands. The specificity of the assay was good. Plasma slightly stimulated steroidogenesis other substances investigated had no effect at physiological concentrations.
