In a medium poorly cationic indirect immunfluorescence of the basement membrane zone normally observed with preparations of sera from patients with pemphigoid has not be seen.

The fluorescence of basement membrane zone takes place if Fe2+ ions are added together with the antibasement zone antibodies. Evidently Fe2+ ions are essential for fixing antibodies.

With the method of indirect fluorescence the C3 component of complement could be observed after fixation of pemphigoid antibodies to the basement membrane zone.

EDTA prevents binding C3 too. Only after adding Fe2+ (not with Ca2+ or Mg2+) complement IF staining is demonstrable. Apparently EDTA causes the loss of fixing antibasement zone antibodies at first. Thereafter complement staining reaction is negative.

Participation of C1 could not be demonstrated using indirect IF staining. The alternate pathway to activation of C3 is discussed.

Durch Versuche in einem kationenarmen Medium gelang es, die normalerweise unter Verwendung von Präparationen aus Pemphigoidseren darstellbare Fluorescenz der Basalmembran bei der indirekten Immunfluorescenz zu löschen.

Wurden den kationenarmen Versuchsansätzen gleichzeitig mit den Antibasalmembran-Antikörpern Ferroinen zugesetzt, fluoreszierte die Basalmembran in der bekannten Weise. Offensichtlich ist Fe2+ für die Bindung der Antikörper essentiell.

Der an der Basalmembran gebundene Komplementfaktor C3 wurde unter Verwendung von Pemphigoidseren mit der indirekten Immunfluorescenz nachgewiesen.

Durch EDTA-Zusätze wurde seine Fixation verhindert. Nur durch Zugabe von Fe2+-, nicht durch Ca2+- und Mg2+-Ionen gelang es, C3 an der Basalmembran darzustellen. Auch hier scheint der Mechanismus über die Fixierung der Antibasalmembran-Antikörper zu verlaufen.

Für eine Beteiligung von C1 wurden mit der indirekten Immunfluorescenz keine Hinweise gefunden. Die Möglichkeit einer Komplementaktivierung über den C3-shunt wird diskutiert.
