Zusammenfassung
Am isolierten, spontanschlagenden Meerschweinchenvorhof wirkt Mg negativ und Ca positiv chronotrop. Im Bereich von 0,4–12,8 mM besteht im Gegensatz zu Ca für Mg eine lineare Beziehung zwischen Schlagintervall und Konzentration. Die pro mMol hervorgerufene chronotrope Wirkung des Mg überwiegt die des Ca um Faktoren zwischen 1,2 und mehr als 5. Für Mg beträgt die Halbwertszeit bis zur Erreichung des maximalen Effektes auf das Schlagintervall etwa 1 min und für Ca circa 2 min. Es wird angenommen, daß für die Änderung des Schlagintervalls die extracelluläre und nicht die intracelluläre Konzentration ausschlaggebend ist. Bei Vorhöfen, die in Lösungen unterschiedlichen Ca-Gehaltes inkubiert wurden, nimmt mit steigendem Ca die Wirkung des Mg ab, während bei Vorhöfen, die mit Lösungen unterschiedlichen Mg-Gehaltes inkubiert waren, die Ca-Wirkung mit steigender Mg-Konzentration beträchtlich zunimmt. Als mögliche Erklärung wird diskutiert, daß die paradoxen Ergebnisse die gegenseitige Beeinflussung von Mg und Ca an verschiedenen „sites” der isolierten Herzpräparation, von denen eine für Änderungen der Spontanfrequenz verantwortlich ist, widerspiegeln.
Summary
In the isolated, spontaneously beating guinea-pig atrium, Mg exerts a negative, and Ca a positive chronotropic effect. Contrary to Ca, for Mg a linear relationship between cycle length and concentration was observed in the range from 0.4 to 12.8 mM. The chronotropic effect produced per mmole Mg exceeded that of Ca by factors between 1.2 and greater than 5. The half-time for the development of the maximal effect on cycle length was about 1 min for Mg and of the order of 2 min for Ca. It is assumed that the extracellular, not the intracellular concentrations of Mg and Ca determine the change in cycle length. In atria incubated at different Ca-levels, the Mg-effect decreased with rising Ca, whereas, in atria incubated at different Mg-levels, the Ca-effect was remarkably augmented by increasing Mg-concentrations. It is discussed, that these paradoxical effects might be the result of the interaction of Mg and Ca at different “sites” of the isolated heart preparation, only one of which is responsible for the change in cycle length.
