Summary
Erythrocyte angiotensinase was obtained by hemolysis and concentrated 5 to 10 fold by sephadex gel filtration. Using the rat bioassay, the enzyme activity was determined as the half time of the degradation rate of angiotensin. Enzymatic activity was characterized with respect to the influence of pH, temperature and various concentrations of different salts. With progressively increasing concentrations of NaCl, KCl and CaCl2, the activity of angiotensinase reaches a maximum and then recedes. Such maxima were not obtained with CdCl2 and Hg2Cl2, rather, a complete inhibition of enzyme activity was noted.
When the enzymatic degradation rate of hypertensin (CIBA) was compared with that of natural angiotensin, the degradation rate of the synthetic substance was approximately 6 times faster.
Zusammenfassung
Die durch Hämolyse aus Schweine-Erythrocyten gewonnene Angiotensinase wurde mit Hilfe eines Rattentests charakterisiert. Dabei wurde die Enzymaktivität aus der Halbwertszeit des Hypertensin-Abbaues ermittelt, die der Enzymaktivität umgekehrt proportional ist. Zur Charakterisierung der Erythrocytenangiotensinase wurde die Abhängigkeit der Enzymaktivität vom pH-Wert, von der Temperatur sowie von der NaCl, KCl, CaCl2, CdCl2 und HgCl2-Konzentration untersucht. Die Angiotensinaseaktivität durchläuft bei Erhöhung der NaCl-KCl-und CaCl2-Konzentrationen ein Maximum, während sie durch CdCl2 und Hg2Cl2 ausschließlich gehemmt wird.
Ein Vergleich der enzymatischen Abbaugeschwindigkeiten des Hypertensin-CIBA und des natürlichen Angiotensins zeigt, daß die synthetische Substanz sechsbis siebenmal schneller inaktiviert wird als das natürliche Präparat.
Mit Hilfe der Sephadex-Gel-Filtration wurde an den Gelen G 50, A 50 und CM C 25 eine Abtrennung des Enzyms vom Hämoglobin und eine Anreicherung um den Faktor 5–10 erzielt. Diese chromatographischen Anreicherungen führten auch zu einer Auftrennung der Aktivität der Erythrocytenangiotensinase. Es konnten zwei Aktivitätsmaxima im Säuleneluat gemessen werden.
