Summary
The treatment of horny tissues with acid solutions of sodium thioglycolate (aNaThG) and osmium tetroxide (Os) is a histochemical method which allows the visualization of the fine structure of different human keratins (callus, nails, hairs) under the electron microscope.
In to this method acid sodium thioglycolate reduces some disulfide bonds of keratins and osmium reacts with free thiol groups. By this way, different keratin patterns can be observed in different horny materials.
In our investigations on hairs and human callus horny cells the characteristicα-keratin pattern could be observed by theaNaThG-Os-Method. Nail keratin however, when prepared under the same conditions as hair, does not show this pattern.
Psoriatic nail keratin is similar to the keratin of normal nails but contains numerous granules of lipid-like material. Furthermore, the intercellular spaces of psoriatic nails are widened and the intercellular coherence is not always preserved.
Zusammenfassung
Die Einwirkung von saurem Natriumthioglycolat (sNaThG) mit anschließender Osmiumtetroxid-Behandlung ist eine geeignete, auf histochemischen Prinzipien beruhende Methode, um die Feinstruktur verschiedener menschlicher Keratine darzustellen. Bei dieser Methode werden die Disulfid-Brücken des Keratins reduktiv gespalten und die dabei entstehenden Thiolfunktionen mit Osmiumtetroxid umgesetzt. DiesNaThG-Os-Methode ist als quasi indirektes Kontrastierungsverfahren spezifischer als die ausschließliche Osmiumbehandlung der Keratine sowie andere direkte, in der elektronenmikroskopischen Cytologie übliche Verfahren.
Mit Hilfe dieser Methode war es möglich, im Haarcortex und in den Hornzellen der menschlichen Schwiele den typischen Filament-Matrix-Komplex des α-Keratins nachzuweisen. Im normalen Nagelkeratin hingegen konnten wir bisher dieses Muster nicht beobachten.
Das psoriatische Nagelkeratin ähnelt dem Keratin des normalen Nagels mit dem Unterschied, daß es zahlreiche sog. Lipoidgranula enthält. Darüber hinaus sind im psoriatischen Nagel die Intercellularräume erweitert und der intercelluläre Zusammenhang nicht immer erhalten.
