Summary
Lymphocytes were cultured in quartz flasks and irradiated by UV light. Time of exposure and moment of application varied. After UV treatment, pulverization of the chromosomes occurred and larger nuclear fragments which might be products of an abnormally proceeding mitosis with chromatin confluence were observed. The usual chromosome aberrations such as breaks, special segments, abnormal chromosomes and polyploidy have been rarely observed, but are found more often in those cultures where pulverization and nuclear fragments are less abundant. A number of mitoses in the irradiated cultures, however, shows no chromosomal aberrations. The ratio of affected to unaffected mitoses is dose-dependent. Chromosome fragmentation can result from UV irradiation occurring at any instance during the time of culture. One finds maximum and minimum amounts of damage when a constant dose is applied at various times.
Zusammenfassung
Lymphocytenkulturen wurden in Quarzgefäßen gezüchtet und mit UV-Licht bestrahlt. Dabei variierten Bestrahlungsdauer und Zeitpunkt der Exposition. Die UV-Strahlung bewirkt eine Pulverisation der Chromosomen. Außerdem treten auch größere Kernfragmente auf, die wir als das Produkt einer abortiv verlaufenen Mitose mit Confluenz des Chromatins werten. Chromosomenaberrationen im üblichen Sinne wie Brüche, Spezialsegmente, abnorme Chromosomen und Polyploidien kommen vereinzelt vor, sind jedoch in den Kulturen gehäuft, in denen Pulverisation und Kerntrümmer selten sind. In einem Teil der Mitosen aus den bestrahlten Kulturen sind keine Chromosomenaberrationen zu erkennen. Das Verhältnis von geschädigten zu ungeschädigten Mitosen ist dosisabhängig. Chromosomenfragmentation kann durch UV-Licht zu jedem Zeitpunkt des Kulturablaufs erzeugt werden. Bei gleichbleibender Dosis erreicht der Anteil geschädigter Mitosen zu bestimmten Zeiten Maximal- und Minimalwerte.
