Summary
In isolated kidneys of Rana ridibunda perfused with solutions free from K+, the net transport (reabsorption) of Na+ is reduced to 30% of the filtered Na+ as against 80–85% in the presence of 2.75 mMol/l of K+. Filtered Ca++ is no longer reabsorbed; the kidneys lose cellular K+. Within the range of 0–2.75 mMol/l of K+ concentrations, these functional changes are fully reversible.
In a preparation perfused with a solution free from K+, the inhibitory action of cardiotonic steroids (convallatoxin 6.4 · 10−6 Mol/l) on Na+ reabsorption is almost completely suspended.
This finding was interpreted as an inactivation of that quota of transport ATPase which is capable of being activated by K+. Since cardiotonic steroids inactivate precisely this quota, they are devoid of any activity in the absence of extracellular K+.
Zusammenfassung
Werden isolierte Nieren von Rana ridibunda mit K+-freien Lösungen perfundiert, vermindert sich der Na+-Netto-Transport (Reabsorption) auf 30% des filtrierten Na+ gegenüber 80–85% bei Anwesenheit von 2,75 mMol/l K+. Vom filtrierten Ca++ wird nichts mehr reabsorbiert, die Nieren verlieren zellständiges K+. Diese Funktionsänderungen sind in Abhängigkeit von der K+-Konzentration im Bereich 0–2,75 mMol/l K+ voll reversibel.
Am K+-frei perfundierten Präparat ist die Hemmwirkung kardiotoner Steroide (Convallatoxin 6,4 · 10−6 Mol/l) auf die Na+-Reabsorption nahezu vollständig aufgehoben.
Die Befunde werden gedeutet als Ausdruck der Inaktivierung des K+-aktivierbaren Anteils der Transport-ATPase. Da kardiotone Steroide eben jenen Anteil inaktivieren, sind sie beim Fehlen extracellulären K+ ohne Wirkung.
