Zusammenfassung
Für die Bestimmung der sauren Spermaphosphatase wird an Hand eines Vergleiches mit Phenylphosphat (Methode nachErdmann-Müller) als Substrat o-Carboxyphenylphosphat empfohlen. Die optimalen Reaktionsbedingungen werden festgestellt. Das Inkubationsmedium ist wie folgt zusammengesetzt: o-Carboxyphenylphosphat 2,5·10−3 M, Essigsäure/Natriumacetatpuffer pH 4,8 0,1 M, Probelösung (bei 2,5 ml-Ansatz). 0,1 ml.
Die Reaktionsgeschwindigkeit und spektrale Empfindlichkeit ist bei Verwendung dieses Substrates wesentlich höher als bei Phenylphosphat. Bei hoher Papiergeschwindigkeit des Schreibers genügen im allgemeinen Spermaverdünnungen von 1∶10 bzw. 1∶100, um meßbare Aktivitäten zu erhalten.
Summary
Estimation of seminal acid phosphatase is carried out with sodium phenylphosphate and o-carboxyphenylphosphate. Comparism indicates more increase of extinction and higher rate of splitting with the aid of o-carboxyphenylphosphate. Dilution of seminal fluid 1∶10 ore 1∶100 is sufficient for estimation. The incubating medium is mixed as following: o-carboxyphenylphosphate 2,5·10−3 M, acetic acid/sodium acetate buffer pH 4.8 0,1 M, seminal fluid (at 2.5 ml test sloution) 0,1 ml.
