Summary
Our results reveal that in psoriasis there is not only an increased proliferation of epidermal cells, but also a disturbance in the metabolism of proliferation with prolonged DNA-synthesis time and mitosis. Using an autoradiographic in vitro double-labelling technique (H3- and C14-thymidine) it was found, that the cell-cycle time was shortened in psoriasis (tc = 114 ± 20 h) as compared with the normal human epidermis (tc = 282 ± 9.6 h). At the same time the DNA-synthesistime (ts = 14.4 ± 1.9 h) and the duration of mitosis (tm = 3.5 ± 0.8 h) were significantly prolonged in comparison with the normal epidermis (ts = 6.6 ± 0.55 h;tm = 1.2 ± 0.17). Special account was taken of diurnal variations in cell-kinetic parameters. At 6 a.m. the H3-thymidine labelling index was increased by a factor of 1.5 in psoriasis. In the normal human epidermis, there were no significant diurnal variations in the H3-thymidine labelling index. The results show that such variations in proliferative parameters are of special interest in investigating DNA-synthesis-time and cell-cycle-time if an in vitro double-labelling technique is applied.
Zusammenfassung
Bei der Psoriasis liegt nach unseren Befunden nicht nur eine gesteigerte Epithelproliferation vor, sondern gleichzeitig eine Störung des Proliferationsstoffwechsels mit Verlängerung der DNS-Synthesephase und des Mitoseablaufs. Autoradiographische in vitro-Doppelmarkierungsversuche (C14- und H3-Thymidin) zeigten bei der Psoriasis eine Verkürzung der Generationszeit (tc = 114 ± 20 Std) im Vergleich zur normalen menschlichen Epidermis (tc = 282 ± 9,6 Std). Die DNS-Synthesezeit (ts = 14,4 ± 1,9 Std) und die Mitosedauer (tm = 3,5 ± 0,8 Std) waren gegenüber der normalen Epidermis signifikant verlängert. Bei der Interpretation der Meßergebnisse wurden die tageszeitlichen Schwankungen der zellkinetischen Parameter geprüft. In den frühen Morgenstunden (6.00 Uhr) war bei der Psoriasis der H3-Thymidin-Markierungsindex um den Faktor 1,5 gesteigert. In der normalen Epidermis variierte der H3-Index mit der Tageszeit nicht signifikant.
