Biochemical Characterization of Connective Tissue Macromolecules Derived from Cutaneous Fibrosarcoma

Summary

Tumor tissue of cutaneous fibrosarcoma was solubilized with salt, acetic acid and salt-extracted after pepsin treatment.

Type III collagen was observed in neutral soluble collagen, 1.5 M and 2.4 M NaCl precipitated fractions after pepsin treatment. Type III collagen fraction precipitated with 1.5 M NaCl eluted in theα 2 region when chromatographed on CM-cellulose without reduction, while the type III collagen partially purified with 1.5 M NaCl eluted between the position ofα 1(I) andβ 12 after reduction and alkylation.

Analysis of amino acid showed the presence of cysteine and the high content of hydroxyproline following CM-chromatography of 1.5 M NaCl precipitated type III collagen fraction.

Acidic glycosaminoglycans were composed of hyaluronic acid, chondroitin sulfate, dermatan sulfate and heparan sulfate, and the first two were major components of fibrosarcoma.

Zusammenfassung

Das Tumorgewebe eines cutanen Fibrosarkoms wurde in NaCl sowie Essigsäure gelöst und unlösliches Kollagen durch entsprechende Pepsinbehandlung in Essigsäure für die Gel-Elektrophorese vorbereitet.

Typ III Kollagen wurde in neutralem löslichen Kollagen, welches in 1.5M und 2.4 M NaCl nach vorhergehender Pepsinbehandlung präzipitierte. Die Typ III-Kollagen-Fraktion, die mit 1.5 M NaCl eluiert wurde, präzipitierte in derα 2-Region bei der Chromatographie mit CM-Cellulose, ohne daß eine Reduktion durchgeführt wurde, während der Typ III Kollagen, welcher partiell gereinigt war und mit 1.5M Kochsalz eluiert wurde zwischenα 1(I) undβ 12 nach Reduktion und Alkylierung präzipitierte. Die Analyse der Aminosäuren zeigte die Anwesenheit von Cystein und einen höheren Gehalt an Hydroxyprolin in der Typ III-Kollagen-Fraktion, die nach der CM-Chromatographie und Präzipitierung mit 1.5 M Kochsalz erhalten wurde. Saure Glykosaminglykane setzen sich aus Hyaluronsäure, Chondroitinsulfat, Dermatansulfat und Heparansulfat zusammen; Hyaluronsäure und Chondroitinsulfat waren die hauptsächlichsten Komponenten des Fibrosarkoms.
