Summary
The occurrence of intracytoplasmic desmosomes in normal, hyperplastic, and hyperkeratotic epithelia, in carcinoma-in-situ and in invasive carcinoma of the human oral cavity is demonstrated by electron microscopy. The mechanism for formation of these structures by invagination, separation and by intracytoplasmic incorporation of plasma membrane-desmosome-complexes are described in various oral epithelia, and other possible mechanisms are discussed. Intracytoplasmic desmosomes may occur in normal and pathological keratinocytes of all layers of the oral epithelium. Their ultrastructure in the peripheral cytoplasm is similar to that of the regular desmosomes on the cell surface. However, as they migrate centripetally, the show signs of degeneration, suggesting dissolution by lysosomal enzyme systems. Various surface membrane alterations involved in the formation of intracytoplasmic desmosomes may lead to a reduction of plasma membrane material and of desmosome structures and to defective intercellular adbesion. The intracytoplasmic incorporation of desmosome structures is a ubiquitous phenomenon exhibited by epithelial keratinocytes under certain physiological or pathological conditions.
Zusammenfassung
Das Vorkommen von intracytoplasmatischen Desmosomen wird in normalen, in hyperplastischen und hyperkeratotischen Epithelien, beim Carcinoma in situ und beim invasiven Plattenepithelcarcinom der Mundhöhle des Menschen elektronenmikroskopisch demonstriert. Der Entstehungsmechanismus durch Invagination, Separation und intracytoplasmatische Inkorporation von Plasmamembran-Desmosom-Komplexen wird in verschiedenen oralen Epithelien aufgezeigt und anderen Entstehungsmöglichkeiten gegenübergestellt. Intracytoplasmatische Desmosomen können in normalen und pathologischen Keratinocyten aller Epithelschichten auftreten. Ihre Ultrastruktur entspricht in der Cytoplasmaperipherie der von regulären Desmosomen der Zelloberfläche; während ihrer zentripetalen Migration lassen sie jedoch Degenerationszeichen erkennen, die auf die Einwirkung von lysosomalen Enzymsystemen zurückgeführt wird. Die Voraussetzungen für das Zustande-kommen von intracytoplasmatischen Desmosomen ist eine Reihe von verschiedenen Alterationen der Zelloberflächen, die mit einer Reduzierung von Plasmamembranmaterial und von Desmosomenstrukturen einhergehen und zu einer Herabsetzung der intercellulären Adhäsivität führen können. Die intracytoplasmatische Inkorporation von desmosomalen Strukturen ist ein ubiquitäres Phänomen epithelialer Keratinocyten, das unter bestimmten physiologischen oder pathologischen Bedingungen manifest werden kann.
