Summary
The juxtaglomerular apparatus in the kidneys of rats was studied by electron microscopy using ruthenium red (block contrasting). The studies showed an intense, partly homogeneous and partly granular-filamentous contrasting of the intercellular spaces of the juxtaglomerular apparatus. The tight junctions and basement membranes in these intercellular spaces were included in the contrasting. The deposit of contrast-rich material was found to extend all the way from the junctions of endothelial cells of the afferent arteriole to the intercellular gaps between cells of the macula densa. Contrasting is interpreted as representing a morphological identification of material transport (tracer effect) in the intercellular spaces of the juxtaglomerular apparatus. Contrasting also provides ultrahistochemical evidence of acid mucosubstances in these intercellular spaces. This may be considered to be proof that acid mucosubstances participate in material transport. When interpreted in terms of transport or carrier mechanism, continuous ruthenium red contrasting of intercellular spaces also shows the course that may be taken by the secretion (renin) produced and given off by the granular epitheloid cells. Thus, secretion discharged into the intercellular spaces may be carried to the nerve fibers and smooth muscle cells and, for final resorption, be transported to the venous or lymphatic systems. The results obtained favor mutual humoral stimulation and regulation of the structural elements of the juxtaglomerular apparatus taking place within the intercellular spaces. Our findings may be considered to provide a morphological basis of a humorally controlled feedback mechanism between the composition of the distal tubular fluid in the macula densa region and the magnitude the preglomerular bloodflow; thus, it corroborates the macula densa theory on the function of the juxtaglomerular apparatus.
Zusammenfassung
Elektronenmikroskopische Untersuchungen mit Rutheniumrot an Nieren von Ratten ergeben eine kontinuierliche intensive Kontrastierung der intercellulären Räume einschließlich dort vorhandener „tight junctions“ und Basalmembranen im Bereich des juxtaglomerulären Apparates von den Junktionen der Endothelien des Vas afferens bis zu den Intercellularfugen zwischen den Macula-densa-Epithelien. Diese Kontrastierung wird als morphologische Kennzeichnung des Stofftransportes (Tracereffekt) und an ihm strukturell (als „Transportschiene“) beteiligter saurer Mucosubstanzen in den intercellulären Räumen des juxtaglomerulären Apparates gedeutet. Bei dieser Deutung zeigt die Kontrastierung die Möglichkeit einer humoralen, in den intercellulären Räumen ablaufenden gegenseitigen Beeinflussung und Regelung der Strukturelemente des juxtaglomerulären Apparates. Die Befunde können als morphologische Grundlage eines humoral gesteuerten Rückkopplungsmechanismus zwischen der Zusammensetzung der distalen Tubulusflüssigkeit im Maculadensa-Bereich und der Größe des präglomerulären Blutstromes betrachtet werden. Sie stützen damit die Macula-densa-Theorie von der Funktionsweise des juxtaglomerulären Apparates.
