Summary
Electron microscopic and topochemical examinations of giant mitochondria with inclusions were performed. The mitochondria were those of muscle tissue from myopathies. The ultrastructural changes consisted of enlargement of mitochondria with an increase of cristae, paracristalline inclusions in the space between the cristae, as well as between the outer and inner surface membranes, and large, round, double-membrane-bound bodies, that contained fragmented cristae with inclusions and concentric membranous bodies. The different structural changes of mitochondria were thought to represent steps of development corresponding to an reaction of adaptation. The formation of giant mitochondria with inclusions is interpreted as a reactive phenomenon and not as primary structural abnormality. The topochemical examinations were performed with enzyme-digestions on ultrathin sections. A digestion-effect was obtained with proteolytic enzymes only, especially with pronase (0.1%/15 minutes). The paracristalline inclusions could be identified as proteins. They did differ from proteo-lipid complexes of cell membranes. The nature of the inclusion-protein as enzyme-protein is discussed.
Zusammenfassung
Elektronenmikroskopische und topochemische Untersuchungen wurden an Riesenmitochondrien mit pathologischen Einlagerungen durchgeführt. Es handelte sich um Mitochondrien aus Muskelgewebe von Myopathien. Die feinstrukturellen Veränderungen waren Vergrößerung der Mitochondrien mit Vermehrung der Cristae, komplexe paracristalline Einlagerungen vorwiegend im Intracristae-Raum aber auch zwischen äußerer und innerer Mitochondrienmembran, und große, doppelmembran-begrenzte Gebilde, die fragmentierte Cristae mit Einlagerungen und konzentrische Membrankörperchen enthielten. Die verschiedenen Strukturveränderungen der Mitochondrien werden als Phasen einer Entwicklung aufgefaßt, die einer Anpassungsreaktion zu entsprechen scheint. Die Bildung von Riesenmitochondrien mit pathologischen Einschlüssen wird als reaktives Phänomen und nicht als primäre Strukturveränderung erklärt. Die topochemischen Untersuchungen wurden mit enzymatischen Verdauungen an ultradünnen Schnitten durchgeführt. Ein positiver Effekt wurde mit proteolytischen Fermenten, besonders Pronase (0,1%/15 min) erzielt. Die komplexen paracristallinen Einlagerungen konnten als Eiweißsubstanzen identifiziert werden. Sie unterscheiden sich von den strukturbildenden Proteid-Lipidkomplexen der Zellmembranen. Die Natur der eingelagerten Eiweißsubstanzen als Enzymeiweiß wird diskutiert.
